Bipyridine phosphodiesterase III inhibitor resulting in positive inotrope effects and vasodilatation

Injection: 1 mg/mL, 10 mL vials

Standard concentrations:
NICU: 100 mcg/mL in D5W
Other areas: 100 mcg/mL; 200 mcg/mL; 400 mcg/mL in D5W;
Inotrope/Vasodilator:
Loading dose: 50 mcg/kg/dose IV over 10 min.
Maintenance: 0.25-1 mcg/kg/min by continuous IV infusion.

*NB: Loading doses in post-op open heart patients require approval from a pediatric intensive care physician.
Bipyridine phosphodiesterase III inhibitor resulting in positive inotrope effects  and vasodilatation.

Contraindicated in obstructive cardiac lesions such as pulmonary and aortic stenosis and coarctation of the aorta.

Side effects include: headache, dysrhythmias, hypotension, hypokalemia, abnormal liver function tests, tremor, thrombocytopenia (lower incidence than amrinone), bronchospasm.

Half-life is about 2 hrs in children with normal renal function.

Half-life is prolonged in patients with renal dysfunction.
