amlodipine__mg PO daily (__mg/kg/dose)
Dosages
Children:
Maximum dose: 10 mg daily.
0.1 to 0.6 mg/kg/dose PO once daily.
Maximum initial dose: 5 mg.Maximum dose: 10 mg daily.
Mechanism of Action
Dihydropyridine calcium channel blocker
Forms Supplied
Tablet: 5 mg
Suspension BCCH: 1 mg/mL
(10 mg tablets are available but not stocked at BCCH).
Comments
Due to long half-life (35-50 hrs) dosage titrations should not be made more frequently than every 3-5 days
Maximum effect on blood pressure may not be seen for 3-4 weeks after therapy begun.
Peripheral edema adverse effect is dose dependent and more like at doses >5mg daily.
Grapefruit may increase plasma concentrations of amlodipine.
